About Danazol

Danazol is an isoxazole derivative of ethisterone (17a-ethinyl testosterone). It has weak progestational and androgenic activities. It is a synthetic androgen (hormone). itused to treat pain and infertility caused by endometriosis (a condition involving the tissue of the uterus). italso used in the treatment of cysts or lumps in the breast.

Indications

Danazol is primarily indicated in conditions like Benign breast disease, Endometriosis, Gynaecomastia, Long term management of hereditary angioedema, Menorrhagia, Premenstrual syndrome, Severe pain and tenderness in benign fibrocystic breast disease, Treatment of endometriosis, and can also be given in adjunctive therapy as an alternative drug of choice in Angioedema, Idiopathic thrombocytopenic purpura.

Contraindication

Danazol is contraindicated in conditions like Vaginal bleeding,Thrombo-embolic complications,Porphyria.

Side Effects

The severe or irreversible adverse effects of Danazol, which give rise to further complications include Hirsutism.Danazol produces potentially life-threatening effects which include Hepatic peliosis, Hepatic dysfunction. which are responsible for the discontinuation of Danazol therapy.The symptomatic adverse reactions produced by Danazol are more or less tolerable and if they become severe, they can be treated symptomatically, these include Sweating, Weight gain, Edema, Hot flushes, Acne, Amenorrhea, Hair growth, Oily skin.

Warnings

Danazol should not be used during pregnancy or lactation. It should be usedwith caution in-patients with migraine headaches, heart, liver or kidney disease, a history of seizures, breast cancer, abnormal bleeding, previous strokes, prostate cancer or any other diseases. Extreme caution is advised for use in children. Pediatric specialist consultation is also advised. Geriatric men are at increased risk of developing enlarged prostate and prostate cancer with the use of androgen-like drugs, so use cautiously.

High Risk Groups

Drug should not be given to Paediatrics, Cardiac / Hypertensive Patients, patients suffering from Kidney dysfunction, Geriatrics, and Neonates.If prescribing authority justifies the benefits of the drug against the possible damages he/she should reevaluate them and consult the reference material and previous studies.
